In the development of fruit flies, how sensitive is the bicoid gradient to the embryo’s transverse length?
Normally, there is a bicoid gradient across the length of the embryo, and upon some threshold bicoid concentration, certain genes are sharply turned on/off, and this distinction is vital for the development of fruit flies.
However, usually the fruit fly embryos do not have identical lengths. If it is true that the material bicoid mRNA is tethered at the anterior end, and the expressed bicoid protein diffuses across the embryo from the anterior source (and the diffusion gradient is identical, regardless of the body length), then would one expect these different embryos to develop body segments at slightly different positions relative to the whole body? Or is there a known error-fixing mechanism to counteract this? This seems so odd since most fruit flies are so uniformly developed in body segments, regardless of the original embryo lengths.
I don’t have the resources to carry out this experiment myself, so I’m posing this question to see if anyone knows the answer. Thanks.
Yes, absolute levels of bicoid will cause differentiation along the AP axis to move forward or back, especially the position of the head fold which is really apparent. Mutants with only 1 functional copy or lines created to carry 3 or 4 copies do see the head fold placed more anterior or posterior. The difference in egg length is quite minimal (maybe 10% variance) compared to having only half or twice as much bicoid and those embryos still develop normally. Throughout the stages of development there are feedback systems in place to ensure that all body parts develop relative to eachother to retain a properly sized and proportioned organism. Many of these mechanisms are not understood as we have yet to find the master genes which control organism size.
